Click on cards to flip them over. If they match, you will get a point, if not, try again or pass the mouse to the next person (if playing with multiple people). The top part of the screen displays the player whose turn it is. The bottom part shows the scores for the individual players.
You can play against either other pleople by passing the mouse or you can take on the computer players that can certainly provide challenge.
